Personality:
Athletic, agile and dangerously unpredictable in a fight, Harley Quinn is a formidable opponent in her own right. She is willing to do anything in order to further the Clown Prince of Crime's goals—no matter the cost to anyone else or even herself. Her energetic, affable and animated personality is enough to make almost anyone drop their guard—a mistake that could easily cost them their life. Deranged and psychotic, Harley Quinn is the poster girl for chaos' terrifying allure.
Through the shock therapy and the Joker's manipulation, her mind and emotions were twisted and she was made into a homicidal, sadistic, child-like psychopath, who would use sexuality and her playful personality to achieve her goals or to entertain herself, usually harming others in the process.
Harley carries almost an innocent persona, wide-eyed and bright, finding humour in otherwise dark situations, much like Joker himself.
Backstory:
Dr. Harleen Quinzel was a psychiatric intern at Arkham Asylum. A model student, in addition to receiving high grades in college, she was also a dedicated gymnast, winning a scholarship to Gotham City University. While researching the lunatics at Arkham, she became fascinated with one particular inmate. Ambitiously volunteering to analyze him, she pleaded with the doctors at Arkham for three months before she could treat him. After he gained her sympathy during their sessions, he seduced her, causing her to fall madly in love with him. The Joker got her to do him a favor, which was giving him a machine gun. Shortly thereafter, Joker's goons broke into asylum, and successfully broke their boss out. Before leaving however, Joker sadistically subjected Harleen to shock therapy. Harleen later chased the Joker down and managed to get him to stop his car. She nearly killed him with a gun, even shooting a man who approached them. The Joker took the gun and taunted her, before slapping her across the face. Harleen wanted to come with him, and he took her to Ace Chemicals. While there, she was told to say the oath, and was asked if she would die for him. She responded in the affirmative, and allowed herself to fall into the vat of chemicals that created the Joker years back. The Joker considered leaving her to die, but promptly changed his mind and jumped in after her, pulling her up to the vat's surface. He then kissed her, and Harleen awakened as the Joker broke down into maniacal laughter.

Quinn's relationship with the Joker is one of the most complex and twisted love affairs in comics: as with all people, the Joker is abusive and manipulative towards Harley, but, just as often, there's evidence of camaraderie, playfulness, and genuine affection towards her. She's the only person who's managed to become intimate on such a long term basis with the Joker.
