Only if they use the command seal that is. There have been some examples of where Assassin class servants came in and killed their masters, or failed doing so.

“Is this the person that attacked you?” Jack’s former teacher lifted up a wallet-sized picture after listening to Jack’s thorough description of his miserable day.
“…yeah. How did you know>”
“She’s been spotted in several random locations all over the world—usually near sites of destruction. We’ve been keeping an eye on her, since her strength is clearly supernatural and she poses a potential risk to the magic society if unwatched. Some have speculated that she’s a Servant, though there’s been no solid proof thus far...” The blond woman sighed. “It would seem the rumors of a hidden Grail War occurring beneath our noses are turning out to be true after all.”
“So then these command spells are real!?” Jack raised his un-gloved hand.
“Looks it to me. Though, the only way to know for sure is to try summoning a Servant.”
“Aaaaagh!” Jack slumped over the counter, scratching his head rapidly in frustration. “But what do I do if it turns out to be real! One encounter with a crazy warrior lady is enough for me!”
His teacher barked a laugh. “For most mages it is an honor to be chosen as a Master.”
“Yeah?” Jack scowled. “Then how about you take my place?”
Sophia Rasputin was a former mercenary—skilled in both war and magecraft—before quitting and becoming a part time lecturer in the Creation Department. Surely she was much more suited to be a Master for the Holy Grail than he.
However, the Russian mage ignored his question and, instead, chose to down her shot in one go. “Isn’t there any wish you want granted?”
“Nothing I’d risk my life for.”
“You can tell that to the Servant that wants to kill you.”
Jack groaned, covering his face in his arms, as his former teacher called for another shot of alcohol. His former teacher would probably be more than a little drunk by the time the night was done. Even so, it was the charmed bartender refilling her glass that wouldn’t be able to remember their conversation at the end of it. Sophia Rasputin was a meticulous magus.

On the hood of Clux’s car, a cat was curled into a resting position as it waited for the owner of the vehicle to real herself. The other humans hadn’t noticed it, too busy going about and following her orders to pay attention to a mere animal. Though, it also might’ve been due to a small charm placed on the feline to make sure that those who looked at it didn’t pay it much attention to it. The ones lacking magical aptitude at least.
When Clux finally arrived, the cat jumped off the hood and made its way to its target.
The cat didn’t open its mouth. However, young female voice was heard coming from the animal nonetheless.“You must be the master of Berserker. Greetings. I am your ally in this Holy Grail War.”

From what I know a servant literally can't, physically can't, kill their master until the command seals are exhausted. The exception being if the servant has a means of bypassing that rule. In virtually every instance of a servant killing their master it's because they trick their master into nulling the contract by using all their command seals, thus no longer being their master.
So, in this situation, Rider physically couln't kill Vita. She literally couldn't do it. That doesn't stop her from swinging a sword at Vita, though.
"If you can't even do your job as a servant then why should I believe anything else you have to say?" Well, this was the moment of truth. If it didn't work she'd just have to make a run for it and call the cops or something.

The dark side currently blends with the shadows so well most of it barely makes an impact so far... *vanishes to sleep*
I actually want the cops to get involved. Like, screw secrecy! Get the world involved! Shani would certainly be happier if she doesn't have to be sneaky.
Gil in FSN isn't a good example because he's not a proper servant at that time. Better example would be F/Z where he had to use Kotomine since he couldn't do anything himself.Didn’t Gil kill Shinji while he was his master in UBW?
